Novotná was widely known for her soprano voice and the roles she performed in the operas of Wolfgang Amadeus Mozart, Richard Strauss, and Leoš Janáček. She began her career at the National Theatre in Prague, where she performed for over a decade before making her debut at the Metropolitan Opera in New York in 1940. Novotná later became a prominent member of the Metropolitan Opera, performing in over 200 productions and earning critical acclaim for her performances.

In addition to her opera career, Novotná also acted in several films throughout the 1930s and 1940s. She appeared in the Hollywood productions The Search (1948) and The Great Caruso (1951) alongside stars such as Mario Lanza and Kathryn Grayson.
