Tripti Mitra started her acting career in the 1940s and gained popularity for her performances in Bengali theater productions. She went on to act in films and television shows, earning critical acclaim for her roles in films like "Jukti Takko Aar Gappo" (1974) and "Balika Badhu" (1967).

Mitra was also a successful stage director and founded her own theater group, "Nandikar", in 1960. She directed numerous plays for the group and was recognized for her contributions to Bengali theater.

In addition to her career as an actor and director, Mitra was also a writer and translator. She translated works of Rabindranath Tagore and other prominent writers into Hindi and Bengali.
